Als große Neuheit wird das bislang bei Musikspielen einzigartige 25 Tasten Keyboard angepriesen. Ebenfalls neu ist der so genannte "Pro Mode", in dem man ein reales Musikinstrument lernen können soll. Als Beispiel hierfür dient eine neue Plastikgitarre mit echten Saiten.

Einer der lästigsten Bugs in Rock Band 3 ist ein Speicherbug, der den Spielstand unbrauchbar macht. Für die PS3 und XBox360 erscheint in Kürze ein Patch, was mit der Wii-Version geschieht ist derzeit unklar.
